Pembroke Street begins by Tennis Court Road and Downing Street in the east and ends at Trumpington Street, across which it continues as Mill Lane. Pembroke College lies along the whole of the south side, which is how the street got its name.

The college frontage

W. D. Caroe worked on the college in the 20th century. His Pitt Building fills the gap between Ivy Court and Alfred Waterhouse's old Master's Lodge, and he lengthened New Court with another staircase beyond the Lodge. An arched stone screen in a late Baroque manner, the Caroe Bridge, ties the two together along the street.

The north side

At the eastern end, on the north side, is the University's New Museums Site. The Department of Chemical Engineering was based there until 2017. Free School Lane leads north off Pembroke Street towards the Whipple Museum. Botolph Lane, a little further north, runs the same way and passes south of Corpus Christi College.

The line of the King's Ditch used to run here, and several historic listed buildings stand along the street.
